Shuttle the ORF insert into another PrecisonShuttle Vector in 30 min
The RapidShuttling kits provide a simple solution for subcloning into various destination vectors. Transferring an insert from one vector to another can be achieved in less than 30 mins. How does it work?
The following components are included:
- Shuttle-ready destination vector (pre-digested & dephosphorylated)
- Sgf I (Asis I) and MluI Restriction Endonuclease (20ul at 1U/ul each)
Or:
Sgf I (Asis I) and RsrII Restriction Endonuclease (20ul at 1U/ul each)
- 10X Rapid Digestion Buffer
- T4 DNA Ligase (for 10 reactions)
- 5X Rapid Ligation Buffer
- Water, nuclease-free

Step 1. Identify the TrueORF clones harboring the desired gene insert (eg. RC200003 for p53)
Step 2. At the TrueORF page, find the restriction sites for subcloning
Step 3. Select the destination vector you wish to transfer the insert into. (View all destination vectors)
Step 4. Select the kit with the correct enzyme combination/destination vector.
