Multiplex microRNA expression measurements with Luminex xMap® technology
In conjunction with catalog or custom Vantage™ microRNA Multiplex Panels, the Vantage™ microRNA Detection Kit provides a fast and simple method for profiling the expression level of up to 100 different microRNAs in a single well.
The Vantage™ microRNA Detection Kit utilizes a simple, hybridization procedure for detection of biotinylated microRNA on the Luminex® instrument. Samples containing microRNA are labeled with a biotin dendrimer using the Vantage™ microRNA Labeling Kit (Cat.# AM100044). The biotinylated samples are incubated with a mixture of different fluorescently dyed xMAP® beads. . Each distinct xMAP® bead is coupled with a unique probe that recognizes a specific microRNA. Probe-coupled bead mixes for specific microRNAs are sold separately as Vantage™ microRNA Multiplex Panels. The beads and sample are incubated at 60ºC allowing the microRNAs present in the sample to hybridize to the specific probes. Following hybridization, the samples are subjected to a high stringency wash to eliminate any non-specific binding. Finally, the samples are incubated with streptavidin-phycoerythrin (SAPE) which binds to the biotinylated microRNA hybridized to the xMAP® bead. The samples are then read on the Luminex® 100/200 or other Luminex-based instruments

The sequences and nomenclature of the mature microRNAs are extracted from The miRBase Sequence Database version 14, released in September 2009, in Sanger Institute in UK (miRBase).
All of the microRNAs are designated by human sequence nomenclature. If the microRNA sequence of your species of interest has greater than 90% homology to the human sequence then the human assay will be able to detect this microRNA in your sample if the normal wash protocol is followed.
All multiplex assays are provided to customers with an additional normalization control assay. Bead 49 detects 5.8S RNA that is ubiquitously expressed in mammalian cells. A signal of 5000-10000 MFI is typically observed when using 500 – 1000 ng of high quality (RIN >8) total RNA.
